Olympic medals 'stolen' as athletes party at nightclub

Rower Alex Partridge was one of two of British Olympic medal winners who had their medals stolen on Wednesday.

By Peter Jeary, NBC News

Updated at 11:22 a.m. ET: LONDON -- Two British Olympians said Wednesday that?their medals were stolen?-- and they've turned to social media in an attempt to get them back.

British rower Alex Partridge and field hockey player Hannah Macleod said their 2012 Olympic bronze medals, along with Partridge?s "Team GB" blazer, disappeared early Wednesday at an upmarket London nightclub. They were among a group of athletes who went to the club?after attending a royal reception at Buckingham Palace to celebrate the British team's success at the 2012 Games.

Under the Twitter name @2012parters, Partridge, 31,?posted a CCTV image with the tweet: ?Does anyone recognize this man as he may be able to help me get my @London2012 bronze medal and @TeamGB blazer back.?

Macleod, 28, tweeted:??My medal was also stolen at the same time. Totally devestated (sic).??

With the name @hannahmacleod6, she also posted the CCTV image, adding, ?This man MIGHT be able to shed some light on the theft ... I'm not after punishment. If you picked up a Bronze Olympic medal that isn't yours pls just send anonymously back to GB hockey-Bisham Abbey.?

In an interview Wednesday night on BBC Radio 5 Live,?Partridge said he initially thought his possessions had been mistakenly taken by another team member. But after looking at a CCTV recording, he said that doesn't appear to be the case.

?Someone?s taken my Olympic blazer with my Olympic bronze medal in it and all I want is to get them both back," Partridge said.

"I'm not after punishment," British Olympic hockey player Hannah Macleod said on Twitter after her medal went missing at a London nightclub.

The full impact of the incident only hit him when he got home, he said.??I suddenly realized my daughter would never see this medal -- what I?d worked so hard for, for 12 years.?

Police?issued an appeal for information into the alleged theft and said the missing jacket, but neither of the medals, had been handed into police Thursday morning.

"My team have spoken to both Olympians who are understandably desperate to be reunited with the medals they have dedicated their careers to win,"?Detective Chief Inspector James Harman said.?

A 29-year-old man had been arrested in connection with the investigation and was being held at a West London police station, police said.

?Phelps, Rupp, Pistorius:?Emotional moments

Dozens of British Olympic and Paralympic medalists had been invited to the palace Tuesday for an event attended by the queen, Prince Philip and the Duchess of Cambridge.

Banners Broker Reviewed ? Does Passive Income Still Exist?

I?m a small business owner with no marketing knowledge. I own a photography and videography studio and most of my business comes from word of mouth and in-industry networking. However, I wanted my business to step it up a notch. I wanted to get my name out their and in the ears of everyone in the city, but I had no idea how to do it? I turned to the radio and television stations, but the prices they asked for were unbelievable! Then, I realized, wait a minute, the internet. Everyone uses the internet! So, began my world wide web business marketing adventure.I spent a few months researching what I was supposed to do to get my website online. After attending a few seminars and speaking with random business owners who had websites I learned about the importance and necessity of SEO and social media.

Check out this cool marketing system that pays 100% commissions here

I myself tried to perform some SEO strategies on my very basic, blog-like website but with no success. So, I decided to hire an SEO company. Every month, they would send me SEO reports, graphs showing my website search engine rank raises banners brokerand all that jazz, but I never saw any increase in profits or met any customers who said they found me because of the internet. So, I decided to try something else. That?s when I found Banners Broker. What Banners Broker does is place banner ads advertising your business all over the internet. You can pay anywhere from $25 to $3000 depending on how much advertising you want. They also seek out the particular demographic you are trying to cater to, find the websites and social media networks they commonly visit and advertise to them. In a sense, Banners Broker subliminally markets your business by stalking them with ads. Sounds creepy, but think about it ? it is beneficial and it is what the big corporations do. For example, have you ever lived a day without hearing or seeing the world Coca-Cola?

You wake up in the morning and check your Facebook ? you see a particular ad. You go to a website to browse and shop around for products ? you see that same ad. You make your daily visit to one of your favorite websites to read an article or two and again, there?s that ad. Eventually, you are going to remember the name of the company ad. Even if you don?t click on the ad, it doesn?t matter, the company name is instilled in your brain. After a while, you become curious. What?s this company all about? Eventually, you go to their website. At least, that?s my idea of what happens. Whether I am right or wrong with these theory, it doesn?t matter, because in just a few weeks after signing up with Banners Broker, I started getting customers who told me they saw my banner ad online and checked out my website. Then, they came to the studio.

What?s also cool about the Banners Broker service is that you can make money using their service. Money that can be used to actually pay for their service. By placing Banners Brokers ads on your site, ads that are relevant to your own business, you can make a little money. The Banners Broker network is like a family of websites that make money by supporting each other.

All in all, Banners Broker combined with SEO and Social Media Internet Marketing, will increase your web traffic and increase your chances for profitability as well as new customer generation. Give them a try. You won?t be disappointed.

ANALYSIS: The Softening Of The GOP Super PACs

After months of relentless attacks on President Obama, the Republican super PACs and independent groups are now going soft. Well, on TV ads at least.

ABC News Political Director Amy Walter notes that Crossroads GPS, the group associated with Karl Rove, released a powerful new ad yesterday featuring the parents of young cancer victim, whom Romney had befriended during his time in the hospital.

And the pro-Romney super PAC, Restore Our Future, went up with a TV spot that featured Iraq veteran Peter Damon talking about Romney visiting him at Walter Reed and the ex-Governor's work "with a charity to build special homes for severely injured veterans." Restore Our Future announced that they plan to spend $17.7 million in 10 states -- all the major battlegrounds plus Michigan -- between October 23 and 29. So, why the personal stuff now?

For the first time in the race, they say, they see an opportunity for Romney to really shrink the gender gap.

Stephen Law, the head of Crossroads, tells ABC, "We've seen a marked shift of women voters toward Romney in the last two weeks, and saw an opportunity to build and deepen that wave." Even so, our own ABC News tracking poll this week showed a 14-point lead for President Obama among women.

Get more pure politics at ABCNews.com/Politics and a lighter take on the news at OTUSNews.com

Romney's strong debate performances have given swing suburban women voters a reason to take a second look at the Massachusetts governor. The question now is whether the second look will turn into an actual vote.

Meanwhile, the pro-Obama super PAC, Priorities USA Action, returned to a familiar theme for their closing-week's ad released yesterday: Romney's record at Bain Capital. It features testimonials from workers at Bain-owned businesses like Donnie Box who says, "This was a booming place. And Mitt Romney and Bain Capital turned it into a junkyard," and Loris Huffman, who adds: "I was suddenly 60 years old. I had no healthcare."

It's one last opportunity for the Democratic group, which has been instrumental painting a picture of Romney as a heartless, corporate raider, to underscore that message.

Lower Back Pain -Women | Cervical dysplasia - abnormal cells ...

Women who have been diagnosed with ovarian cancer often say that they experienced mild to heavy lower back pain before their treatment.

The back pain occurs in the lower back and often feels like a dull ache. Some women describe it as feeling like labor pains.

My back ached a lot. At first it was occasional like when I did dishes or the laundry, but then it began to ache when I was sitting, doing nothing. I knew something had to be wrong. I went to my doctor and later found I had stage III ovarian cancer.
Candace, Seattle, WA

Any symptom the lasts more than a week or two really needs to be evaluated by a doctor. Keep in mind that lower back pain may be a sign of PMS for some women. However, chronic lower back pain should not go untreated.

Venice winner "Pieta" director a soft-spoken "monster"

YANGPYEONG, South Korea (Reuters) - The near-death of an actress in an accident while filming nearly ended director Kim Ki-duk's career four years ago, but after making "Pieta," which took best picture at this year's Venice film festival, he is now South Korea's most feted auteur.

The incident, in which an actress playing a character who hanged herself fainted with the rope around her neck and was cut down by Kim himself, shook Kim so badly it changed his views on mortality.

Hit with a subsequent wave of staff departures, he retreated from the world to live in a rough wooden shack he built himself about an hour outside of Seoul.

"For the past two to three years, I believed there was no value in my life any more and did not make any movies," said the soft-spoken 52-year old, his hair tied back and wearing shabby chestnut-colored Korean traditional clothes.

"I hated everything. Then I thought life was way too long," Kim said of his self-imposed exile.

But the working-class Kim, who has been tagged by some feminist critics as "all evil, no good," a misogynist or even a psychopath, picked himself up to make "Arirang" in 2011 and then the ultra-violent "Pieta."

"Pieta" depicts the relationship between a heartless loan shark and a middle-aged woman who says she is his mother. Although critics say it is less brutal than many of Kim's other films it still features mutilation, sexual violence and cannibalism as the loan shark feeds the woman his own flesh and rapes her.

The movie, which Kim said he made as a comment on capitalism in Korea, scooped the Golden Lion award at Venice, where one critic termed it "intense."

"I felt that people still overlook the essence and that they are overly judgmental about a certain scene or a person," he said when asked about the frequent negative comments about his movies or his personality.

The movie is shot in the Cheonggye district of central Seoul, once a maze of factories and sweat shops that has now been largely bulldozed. The newly shiny urban area in the film is a place where people are so poor they are prepared to barter their body parts for cash.

"Extreme capitalism is a global phenomenon... Pieta asks its first question about these issues and secondly, it raises the problem of how money dissolves family and human relations," Kim said of his 18th film, the first South Korean film to win a major international award.

MIDDLE SCHOOL EDUCATION, FACTORY WORK

Kim was born in 1960 and his education went only as far as middle school. He then worked in the factories that helped power South Korea into an industrial nation before giving it all up to go to Paris to draw.

Back in Korea, the self-taught director debuted with his low-budget movie "Crocodile" in 1996. He has remained an outsider and a minority taste ever since, describing himself as a "monster who grew on his inferiority complex".

"(It) is an element that makes me train myself harder and grow bigger. The word 'monster' sounds as if it's something enormous, unidentifiable and predatory, but monster means monster. I don't think it's that bad," Kim said.

Korea's critics appear to be softening. They note that his films appear to have become more accessible and that something "Kiduk-esque" has faded from his movies.

"While he has sought aesthetic aspects before, now there are signs that he's trying to shift to moral aspects," said Lee Taek-Gwang, a cultural critic and a professor of English Literature at Kyunghee University.

Kim appears willing to accept the risk he may become more popular, but says it is not something he's actively seeking.

"Well, I think it's something I should reflect on if many think I've become popular or audience-friendly," Kim said when asked whether his work had become more approachable.

"Even though I'm not saying they are wrong, I feel I still have cruel scenes and something gruesome (in my films)."

The Note's Must-Reads for Tuesday October 23, 2012

The Note's Must-Reads are a round-up of today's political headlines and stories from ABC News and the top U.S. newspapers. Posted Monday through Friday right here at www.abcnews.com

Compiled by ABC News' Carrie Halperin, Jayce Henderson and Amanda VanAllen

TAKEAWAYS ABC News' Abby Phillip: " Who Won? Political Analysts on the Final Debate" The final presidential debate on foreign policy appeared to be a win for President Obama, but a successful effort by Republican nominee Mitt Romney to return the conversation to the economy, ABC News political analysts said. "The president won tonight," said ABC News political analyst Matthew Dowd. "Here's why: I think they both came in with different agendas, but the president was more successful." LINK

Politico's Maggie Haberman: " Presidential debate 2012: 7 takeaways" The curtain has fallen on the final debate of the 2012 cycle, yet one question remains unanswered - will it change anything? President Obama was on the attack, Mitt Romney modulated toward the center, and Bob Schieffer played it low-key in the moderator's seat in the Lynn University face-off in Boca Raton, Fla. LINK

The Wall Street Journal's Janet Hook: " Rivals Seek Points by Going Off-Topic" The sponsors promised a foreign-policy debate, but the candidates didn't stick to the script. President Barack Obama and Republican challenger Mitt Romney ranged far from international affairs, wading deep into the economic issues that are voters' top concern. The path was dictated more by the candidates' political needs than the agenda of debate organizers. LINK

REVERSING ROLES ABC News' Russell Goldman: " Whoppers, Bayonets and Zingers in Final Presidential Debate" President Obama was on the offensive during the third and final presidential debate tonight, hitting Mitt Romney for shifting on foreign policy positions and pointedly telling Romney, "Every time you've offered an opinion you've been wrong." Romney agreed with Obama during much of the debate that concentrated on foreign policy, but had sharp words for the president on the Middle East saying the president "wasted these four years" by failing to stop Iran's nuclear program and allowing the Middle East to descend into "tumult." LINK

The Boston Globe's Glen Johnson: " Obama gives his strongest debate performance" President Obama dissected Mitt Romney's foreign policy views on Monday, suggesting the former businessman lacked the familiarity, consistency, and forward-looking approach to be an effective president. It was perhaps the boldest statement of a night that saw Obama turn in his strongest debate performance against Romney. If there was one downside, it was on the topic - foreign affairs - that polls have consistently shown ranks well behind job creation and economic recovery as most important on voters' minds. LINK

The Hill's Justin Sink and Amie Parnes: " Obama hits Romney hard in debate" President Obama and Mitt Romney took each other on in the final presidential debate of the 2012 campaign Monday night, fighting to prove their credentials as the right leader for America on the world stage. Obama battled aggressively, repeatedly throwing darts at Romney and seeking to portray his opponent as "wrong and reckless" on foreign policy. LINK

The Los Angeles Times' David Lauter and Christi Parsons: " Obama reverses roles, comes out swinging at Romney in final debate" President Obama and Republican nominee Mitt Romney made their closing appeals to voters in a final debate that featured an unusual reversal of strategy - Obama adopted the typical challenger's stance of underscoring differences, while Romney repeatedly sought to mute them. LINK

The New York Daily News' " Presidential debate: Determined Obama goes on attack, painting Mitt Romney as reckless and inconsistent in world affairs" President Obama lacerated Mitt Romney Monday during their third and final debate Monday, repeatedly painting his Republican rival as wrong, reckless and inconsistent in world affairs. Romney accused Obama of being ineffective on the global stage - but in a surprise, he soft-pedaled several of his differences with Obama and even agreed with the President on a host of policies. LINK

The Washington Post's Dan Balz and David Nakamura: " Obama keeps Romney on his heels in last debate" President Obama and Mitt Romney clashed repeatedly over foreign policy here Monday night, with the president arguing assertively that Romney has lacked the consistency or clarity of vision to lead the country while the Republican nominee charged that Obama has been weak and ineffective in the face of growing turmoil in the world. The two candidates differed most sharply over the president's handling of the uprisings in the Middle East, his efforts to prevent Iran from obtaining nuclear weapons and his treatment of Israel. LINK

FOREIGN POLICY The Washington Times' Stephen Dinan: " Obama, Romney tangle on al Qaeda, foreign policy in final presidential debate" Mitt Romney accused President Obama of failing to protect the military from budget cuts and squandering U.S. leadership in the Middle East, leaving America standing by as al Qaeda has surged to become active in a dozen countries, as the two men faced off Monday night in their final debate. LINK

The New York Times' Peter Baker and Helene Cooper: " Obama and Romney Bristle From Start Over Foreign Policy" President Obama and Mitt Romney wrapped up a series of defining debates on Monday night with a bristling exchange over America's place in the world as each sought to portray the other as an unreliable commander in chief in a dangerous era. Picking up where he left off in last week's debate, Mr. Obama went on offense from the start, lacerating his challenger for articulating a set of "wrong and reckless" policies that he called incoherent. LINK

Bloomberg's Julie Hirschfeld Davis and Indira A.R. Lakshmanan: " Obama Cites Foreign Policy Wins as Romney Mostly Agrees" President Barack Obama kept Mitt Romney on the defensive on foreign policy in their final debate before the election, as his Republican rival worked to criticize the incumbent's leadership while endorsing most major actions he has taken. The president sought to portray the former Massachusetts governor as ill-prepared to assume the mantle of commander in chief, branding his statements on international challenges as "all over the map" and his positions "wrong and reckless." LINK

GOP USA Today's Susan Davis: " GOP has firm grasp on House, but Dems Won't fold" Control of the White House and the U.S. Senate is up for grabs on Election Day, but as certain as anything is in politics, it's safe to bet that Republicans will continue to control the U.S. House in the next Congress - and maybe well beyond. Republicans control the House, and although Democrats have many good prospects to chip away at their majority, the nation's top election forecasters are decisively pessimistic about Democrats' chances of gaining the 25 seats they need to take control of the chamber. Not just for kids: The most romantic Disney vacations offer grown-ups a place to play

2 hrs.

On a recent vacation, business manager Lauren O?Donnell?s boyfriend set the mood with a private boat ride. Fireworks lit up the sky, and he popped the question?at Disney World.

?I texted my friend to let him know I?m engaged,? says O?Donnell, who works at American Express Publishing, ?and learned he went to Disney for his one-year anniversary last year.?

Slideshow: See where the most romantic Disney vacations are

Grown-ups like to play at the Happiest Place on Earth, too, and romance has become big business for Disney. Over the past two decades, Disney World alone has hosted more than 45,000 weddings, and countless couples have vacationed at the company?s properties across the U.S.

?I think that Disney has this intangible magic that envelops people, if they allow it to,? says Deb Wills, whose Disney news and travel-planning site AllEars.net has a devoted following of 145,000 weekly subscribers. ?You can shut out the real world; you can suspend reality.?

But disappearing into that kind of theme-park love cocoon isn't always easy.

?Disney is Disney,? says Wills. ?There are lots and lots of families.? So start by going where the children aren't, such as a resort hotel during the daytime, when most guests are on the park rides. Or a cruise ship like the?Disney Wonder?and?Disney Magic,?on which couples? spa treatment rooms pair a massage with champagne on a veranda with sea views.

Inside the theme parks, privacy is catch-as-catch-can on little-walked trails and quiet culs-de-sac (ahem, try wandering to the back of Paradise Pier at California Adventure).

For a more guaranteed dose of romance, book a table at Be Our Guest restaurant, opening in?November at the newly expanded Fantasyland. It?s the first Magic Kingdom restaurant ever to serve wine and beer with dinner, and the d?cor is also creating buzz: a domed dining room with views of falling ?snow? that echo the ballroom scene from "Beauty and the Beast."

We've rounded up more couple-friendly Disney activities, each different in mood and expense. If you aren't swooning by the end, you may be better off at Six Flags.

In Amendment 4 Campaign, Real Estate Agents Raise $4 Million ...

John Sebree of the Florida Association of Realtors said Amendment 4 will help stimulate Florida?s economy. (Photo from?Florida Association of Realtors video.)

By Ashley Lopez
Florida Center for Investigative Reporting

Real estate agents have raised $4 million to support Amendment 4, which if passed will enact property tax limitations that could shrink the revenues Florida counties and municipalities need for services.

Amendment 4 would cap the assessed values of homes and lower property taxes for investors, businesses, and owners of rental properties and second homes. It will also increase the number of people eligible for homestead exemptions. In their efforts to pass Amendment 4 on Nov. 6, real estate agents have raised more money than any other campaign for or against a ballot measure this year.

These real estate agents say the amendment will improve the state?s faltering housing market, which will in turn boost the state economy.

The primary opponents of Amendment 4 are Florida?s local counties and cities, which are prohibited from spending money to influence the outcome of ballot measures. The leaders of these local governments, concerned they will not be able to fund services properly if Amendment 4 passes, have resorted to passing resolutions opposing the ballot measure ? one of the few actions local governments can take against Amendment 4.

Lakeland, Gainesville, Marianna, North Port, and Palm Beach are among the Florida municipalities that have announced opposition through these resolutions.

Public officials in Florida fear Amendment 4 will raise taxes for homeowners who live in their homes year-round and lower taxes for investors and snowbirds ? all while resulting in substantial cuts to local government services such as police, fire and waste management.

Bill Horne, Clearwater?s city manager, estimates that Amendment 4 would take more than $414,000 out of his city?s coffers in 2013.

?I can?t tell you where I am going to get savings,? Horne said. ?Our only options are to reduce staffing and services.?

Supporters of the amendment, however, believe it will jumpstart the state?s housing market, which will in turn increase tax revenues.

?We may take $40 million out of local government by passing something that benefits a taxpayer, but the true impact of it will be a positive because its spurs economic growth,? said John Sebree, senior vice president of public policy of the Florida Association of Realtors. ?It creates jobs. It allows people to spend more money.?

Florida TaxWatch, an independent public policy group in Tallahassee that has not taken a position on the ballot measure, estimated that Amendment 4 would create as many as 383,810 additional home sales over the next decade.

How Amendment 4 Works

If passed, Amendment 4 would cap assessed home values, lower property taxes for investors, businesses, and owners of rental properties and second homes, as well as increase the number of people eligible for homestead exemptions.

Ken Small, a finance expert at the Florida League of Cities, said the impact of these new rules would devastate local governments, because some real estate owners will eventually pay little or nothing in property taxes.

?If the economy picks back up again,? Small explained, ?it will take probably decades before the taxable value of counties and cities gets back to a level where they had been, because [this] cap will hold it down.?

While some residents will get new property tax breaks, Small said, others will have to pay more because local governments will be forced to raise millage rates to pay for basic services or find ways to decrease government services.

Cities such as Clearwater, in Pinellas County, have already made cuts in the past few years as the state?s plummeting housing market and struggling economy pushed down tax revenues. These cities have only begun to recover, and Horne and Small agreed that Amendment 4 would likely set that recovery back.

?It?s a frustration when trying to come up with a way to deal with these cuts,? said Horne, Clearwater?s city manager. ?We think things are getting better moving forward, but with these amendments, we are going to go back to where we were.?

In Pinecrest, a wealthy suburb in Miami-Dade County, Village Manager Yocelyn Galiano Gomez estimates that Amendment 4 would create a $1.9 million tax loss over a five-year period.

While worried about how Amendment 4 will affect Pinecrest, she is even more concerned about other, more urban municipalities in Miami-Dade County.

?Those cities with larger commercial areas, such as Doral and Hialeah, will stand to lose much more,? Gomez said.

The Big Push

Real estate agents say Amendment 4 will revive the state?s struggling housing market by giving people an incentive to purchase homes.

The Florida Association of Realtors and the National Association of Realtors together have raised $4 million on a pro-Amendment 4 campaign. The ballot measure, if passed, would likely mean better business for real estate agents and property investors.

Mailers sent statewide and paid for by this campaign are promising more jobs, increased home sales and accelerated economic growth if Amendment 4 passes.

Sebree, of the Florida Association of Realtors, said cities would only have to cut services if they plan to spend more in the future than they are spending now.

?I think local governments want to always be able to increase their spending based on a formula,? Sebree said. ?You know, they want to spend more this year than they did last year ? This isn?t going to cut that ? it just puts a cap on assessment increases.?

Conflicting Studies

The net effect of Amendment 4 is debatable because reports and studies conflict.

A June 2012 Florida TaxWatch report estimated Amendment 4 would create 19,483 private-sector jobs over the next decade. A March 2010 report from the same group, however, claimed that tax assessment caps would deliver ?fewer jobs? because they are an ?impediment to a vibrant economy.?

The state?s Office of Economic and Demographic Research estimates that statewide revenue cuts would be $118 million the first year and swell to $321 million in the third year if millage rates remain the same.

Cragin Mosteller, a spokeswoman for the Florida Association of Counties, said the boon to real estate agents would end up hurting others in the state. She also said county governments are concerned that it will cap the ability for communities to invest in themselves.

?I think the Realtors are trying to sell it as a first-time homebuyer tax cut, but that?s really not fair,? Mosteller said. ?It?s a tax cut for our snowbirds and investment property owners ? you know, those guys that flip real estate ? but it will also be a cost shift to Florida residents.?

Turbo switch of calcium pump in biological cells discovered

ScienceDaily (Oct. 21, 2012) ? A Danish-British research team has discovered a turbo switch in the vital calcium pump in our body's cells. In studies at the X-ray source DORIS at Deutsches Elektronen-Synchrotorn DESY in Hamburg and the European Synchrotron Radiation Facility ESRF in Grenoble the team discovered that the on-off switch of the pump has a previously unknown third position, which switches the pump into a turbo gear.

The group of Henning Tidow from Aarhus University and Lisbeth Poulsen from the University of Copenhagen published its studies in the British journal Nature. "The discovery not only improves our understanding of a fundamental mechanism in the biology of all higher organisms, but could one day allow for better treatment of certain diseases in which the calcium balance is disturbed," says Tidow. The researchers used the measuring station of the European Molecular Biology Laboratory EMBL at DORIS.

The element calcium plays a central role in many processes of life, such as cell division, the day-night cycle and the communication of cells. The decisive factor is a gradient in the calcium concentration, which is normally high outside the cell and low inside it. This gradient is maintained among others through a calcium pump, which occurs in all higher organisms (eukaryotes) -- from the nettle to the blue whale. For example, under stress the calcium concentration in the cell increases and triggers a corresponding reaction. Afterwards, the concentration must be lowered again.

"The calcium transport from the cell requires a lot of energy. It is therefore important that the pump is activated only when needed," explains Poulsen. The pump -- known as PMCA (plasma-membrane calcium-ATPase) -- thus has a switch, which is actuated by the protein calmodulin. When calcium binds to calmodulin, the latter changes its shape so that it can dock onto a binding site of the cell's calcium pump, thereby activating the pump. When the calcium concentration in the cell increases, more and more pumps are thus switched on.

The researchers led by Tidow viewed the entire switching complex with X-rays to reveal its molecular structure. They chose the switching complex from cells of the plant thale cress (Arabidopsis thaliana), studying it first in crystal form and then in solution, which is closer to the natural environment of the molecule. "Based on this analysis, we were able to create a detailed three-dimensional model of the region of the calcium pump that interacts with calmodulin," says Tidow. "To our great surprise, we found that the calcium pump has two binding sites for calmodulin and not just one as previously thought."

The switching complex thus consists of a dumbbell-like structure with two calmodulin binding sites. To determine whether the second site has a biological significance, the researchers tested pumps in which they had disabled one switch. Indeed, these pumps could not run at full power. "Our results show that the calcium pump is controlled in three steps," explains Poulsen. "It is switched off when no calmodulin is bound to the switching complex. The pump is running at medium speed as soon as one binding site is occupied, and at full speed when calmodulin is bound to both sites."

The pump is thus activated step by step, depending on how much calcium is present in the cell. When the calcium concentration increases, the pump first operates in an energy-efficient way at moderate speed. If the calcium threatens to reach an amount that is dangerous for the cell, the pump changes into the turbo gear, which enables it to very quickly reduce the concentration.

Bioinformatics analyses revealed that this double switch occurs not only in all plant species, but in general in all cells with a nucleus (eukaryotes). "This study underscores the strength of integrating structural biology in interdisciplinary research," underlines Poul Nissen of Aarhus University. As the next step, the researchers aim to decipher the structure of the entire calcium pump.

Yahoo CEO's comeback plan hones in on technology, not media

SAN FRANCISCO (Reuters) - Marissa Mayer, who earned a reputation for decisive action and intensity during her 13-year stint at Google Inc, has spent her first months as Yahoo Inc CEO quietly moving the Internet pioneer back to its roots in technology.

Long torn between whether it should focus on media content or on tools and technologies, Yahoo under Mayer is being positioned firmly in the latter camp, according to sources inside and outside the company.

Her hires, acquisition musings, and other early moves hint at an ambitious, technology-driven comeback plan designed to revitalize aging but well-trafficked properties such as Yahoo Mail, Yahoo Finance and Yahoo Sports.

Yahoo has been criticized for allowing these sites to stagnate - they look very much like they did five years ago, and do not have many bells and whistles to encourage users to spend more time on them.

Mayer, 37, wants to make Yahoo's properties much more interactive, on PCs and on mobile devices, using social media tools to personalize the user experience and new technology to boost advertising sales. Her well-known focus on user design is expected to result in a simpler, less-cluttered email and home page, one source said.

Yahoo declined to comment for this article. Mayer, who gave birth to her first child weeks ago, will unveil details of her comeback plan when Yahoo reports quarterly results on Monday.

Mayer's focus on technology in many ways reverses a course set by her predecessors, who had concentrated on media content deals, such as those that gave prime billing to Walt Disney Co's ABC News or CNBC, or to bring an original program starring actor Tom Hanks to its website.

The new strategy is not without risks: it positions Yahoo squarely against Facebook Inc and Google. It also risks alienating a large, media-focused contingent that is already weakened by the departure of Ross Levinsohn, who had championed a media-centric approach when he was interim CEO before Mayer's arrival in July.

Mayer has been meeting with Internet gurus including AOL Inc CEO Tim Armstrong, another ex-Googler; Silicon Valley lawyer Larry Sonsini; and Wall Street investment bankers, according to people familiar with the matter.

Bankers have pitched Mayer and her team on a slew of potential acquisitions, and they appeared to show interest in restaurant reservation site OpenTable Inc and advertising technology companies PubMatic, Turn and Millennial Media, one of the people said.

Caterva, a small start-up whose technology analyzes social media activity, has also been in low-level talks with Yahoo, said another source familiar with the situation.

OpenTable and PubMatic declined comment. Millennial Media and Caterva did not respond to requests for comment.

With more than $2 billion in cash and short-term securities, Yahoo has the money to acquire engineering talent or bolt-on services. Two types of deals are under consideration: companies that will increase user engagement, including on mobile, and those that will boost advertising returns, source said.

"What they've signaled so far is that the deals will be more niche in nature, smaller deals that maybe have a lot of promise," said Ken Allen, a director at Blackstone Advisory Partners.

TALENT HUNT

Many industry insiders believe Mayer is Yahoo's final hope for reversing a years-long decline from the pinnacle it once attained as the leading gateway to the Internet. Four of her predecessors have tried in vain to right the ship - Yahoo's market value of $19 billion, is less than half its $44 billion value in 2005.

Mayer, who earned a masters degree in computer science from Stanford University specializing in artificial intelligence, has moved quickly on the personnel front, shelling out rich pay packages to attract ex-colleagues from Google and elsewhere.

She brought in ad technology systems guru Henrique de Castro as chief operating officer; a new finance chief in Ken Goldman, who also has tech chops, to replace Tim Morse; and Jacqueline Reese to assume the dual role of hiring and acquisitions, suggesting the start of a train of "acqui-hires" or buying small companies for their engineering talent.

"She's spending almost all her time with the product folks. She's spending it on technology. She's talking about engineering hires," a person close to Yahoo said about Mayer's early days.

Yahoo's advertising technology products, headed for the auction block before Mayer's arrival, are back in favor. De Castro, her highest-profile hire, is known for a deep-understanding of the complex advertising landscape, where dozens of businesses and technology providers are interlinked.

Mayer has also shown an interest in the company's ad tech platform, including Right Media, an automated exchange that allows marketers to blast ads across a network of websites.

The group has been a long-standing source of division among Yahoo's management, including with Levinsohn, who was keen on divesting the unit, according to two sources close to the matter. But shortly after Mayer's arrival, Yahoo told AdAge that it had no intention of selling Right Media.

Yahoo's advertising salesforce, responsible for signing splashy home-page ad deals and premium marketing campaigns, has received scant attention from the new CEO, say people close to the company. Michael Barrett, Yahoo's chief revenue officer hired by Levinsohn shortly before Mayer's arrival, recently announced his resignation, according to a source familiar with the matter.

FOCUS ON MOBILE

Roughly 700 million users visit a Yahoo website every month - putting it in the top ranks globally. But the amount of activity people engage in on many sites is steadily declining, and its smartphone offerings are deemed lackluster.

"The largest change is to be deadly serious about mobile," said a former Yahoo manager who remains in touch with people at the company.

Yahoo faces tough competition from Facebook and Google, two companies that have taken consumers' time, engineering talent and market value from Yahoo. They are also trying to make the transition to mobile, but it has been difficult.

Some say the direction signaled by Mayer is not so different than strategies espoused by previous CEOs that Yahoo has consistently struggled to implement. A fragmented culture in which short-term finances usually trump product plans is to blame, according to those who know the company.

The recent departure of CFO Tim Morse could signal a change in approach, said several former Yahoo employees.

Morse was considered the force behind Chinese e-commerce company Alibaba Group and Yahoo's $7.6 billion deal over the summer, which saw Yahoo sell about half of its 40 percent stake in Alibaba after years of wrangling over terms.

But now Yahoo's Asian partners, including Yahoo Japan Corp, are not on the front burner for Mayer, one source familiar with the situation said.

Whether Wall Street has the patience for yet another Yahoo revival plan remains to be seen.

"Every CEO needs time to have their full vision articulated and understood," said Dan Rosensweig, a former Yahoo chief operating officer, who now serves as CEO of online textbook rental company Chegg.com. "To count Yahoo out would be an enormous mistake, because the users have not counted Yahoo out," he said. "It's not like MySpace, where all the users went away."

Web of relationships aids success in kids, foundation co-founder says

GENEVA ??When school officials want to teach youths about the dangers of drugs, they might do a presentation. International child-youth advocate Derek Peterson dismissed that as ineffective.

?An in-service without bonding is a ?drive-by,? ? Peterson said this week during a two-day workshop on how to improve student performance and reduce at-risk behaviors. ?The adults come out, do a workshop and say, ?We did what we could do to stop drugs.? No, we didn?t.?

Peterson, a co-founder of Changing Children?s Worlds Foundation, a charity based in Geneva that focuses on violence prevention, said the way to help youth succeed is to bond with them first. The workshop was at the group?s headquarters at 411 Stevens St.

Peterson said research shows teenagers who sit through a presentation do not change their behavior. But with bonding, data shows a change toward success.

?This is so controversial because it takes a lot of time,? Peterson told a small group attending the workshop. ?But when you take that time ? I swear to God, it leads to change.?

Peterson uses the imagery of strings in a web to illustrate how a children?s relationships support and protect them while they are developing. The more strings in the web, the more support and improvement in development. Less support, the more likely youths will engage in dangerous or negative behavior that can affect them all their lives, Peterson said.

Peterson has worked with community groups across Arizona. Through hands-on workshops, Peterson said people saw resources they could offer their youth and collected them in a book, ?Helping Kids Succeed Arizona Style.?

Kimberly Svevo-Cianci, president and executive director of Changing Children?s Worlds Foundation, said she wants a similar effort in Kane County.

Nate Micatka, 16, a junior at Geneva High School, said he was glad he came to the workshop.

?I learned that everybody has a web, and they need to be supported by five adult figures,? Micatka said. ?Parents, teachers ? or just someone who can be there for you.?

Two Kane County sheriff?s deputies, Krysta Kaus, school resource officer in Burlington, and patrol officer Andrew Biddle attended.

O2 UK begins rollout of Jelly Bean to Galaxy S3, Vodafone to follow

Galaxy S3

Yesterday, Three UK began to send out the Andorid 4.1 (Jelly Bean) update to Samsung Galaxy S3 phones, and now we have two more British carriers following closely -- Vodafone and O2. O2's rollout will begin today, it says, while Vodafone customers will have to wait until Monday.

Again, we're pleasantly surprised at how fast the British and European carriers have been able to get the updates to their devices and hope those on the American side are paying attention. Both updates will be a gradual rollout and are available via OTA over WiFi or via Kies.

If you have questions about a specific carrier's implementation, hit up the source links. We'd love to hear from you if you're on one of these carriers and update your Galaxy S3.

Motte goes 2, makes Cardinals' wait worth it

ST. LOUIS (AP) ? Jason Motte had a lot more time than usual to get ready, learning the plan before the grounds crew took the tarp off the field. It was nice to get some advance notice before going for the first two-inning save of his career under October pressure.

The San Francisco Giants went six up and six down against the St. Louis Cardinals' hard-throwing closer, thrust into an extended appearance by a rain delay that lasted longer than Game 3 of the NLCS. Motte needed just 19 pitches to complete a 3-1 victory that was worth the wait Wednesday night, putting the defending World Series champions up 2-1 in the series against the 2010 World Series titlists.

"I've never had a situation where I've known that far ahead of time," said Motte, who has saved both of St. Louis' victories in the NLCS and has three saves in the postseason. "You know, you're usually watching the game and you prepare yourself that way.

"I went to the bullpen a little early and got my arm loose and I was able to get the job done."

Rookie Matt Carpenter hit a two-run homer in his first at-bat of the NLCS after subbing for the injured Carlos Beltran and the Cardinals chased Matt Cain just before a 3?-hour rain delay in the seventh inning.

Beltran strained his left knee running out a double-play ball in the first inning and manager Mike Matheny said he was day to day. He's had issues with the knee throughout the season, but played in 151 games and had 619 at-bats, his most since 2008.

Cardinals pitchers made the lead stand up, beginning with Kyle Lohse, who worked around a season-worst five walks in 5 2-3 innings. The 16-game winner is 2-1 with a 1.96 ERA this postseason after escaping several jams, and has allowed two or fewer runs in all three of his starts.

"I made up a word. Grindy, very grindy," Lohse said. "Just one of those days where I knew warming up it was not quite coming out the way I wanted it to."

Setup man Mitchell Boggs pitched earlier than usual, too, bailing out Edward Mujica. Boggs struck out Hunter Pence and Brandon Belt with two on to end the seventh as the storm approached.

Motte made it a rewarding finish for what remained of a sellout crowd of 45,850 ? perhaps a third ? in a game that lasted 3 hours, 2 minutes. He was ready to go for Game 4, too.

"I feel great," Motte said. "This time of the year, you're ready to go every single day."

Giants second baseman Marco Scutaro had two hits and a clean game in the field, two days after Matt Holliday rammed into him breaking up a double play. Manager Bruce Bochy had said there would be no retaliation, and Game 3 was collision and controversy free.

"He was determined to play and made a pretty good recovery," Bochy said of Scutaro. "He played well, got some big hits. It says a lot about his makeup, and how tough he is."

Cain lost for the second time this postseason, giving up three runs on five hits in 6 1-3 innings. The Giants, who entered the game batting just .217 in the postseason, were 0 for 7 with runners in scoring position and stranded 11 runners.

Pence, the Giants' fifth-place hitter, also grounded into a double play with runners on first and third in the third and grounded into a force play with a man on to end the fifth.

Except for a hanging slider to Carpenter, Cain was happy with his outing. He's allowed three runs in all three postseason starts.

"I was locating both sides of the plate," Cain said. "Throwing everything for strikes, and getting ahead of guys. This is the best I've felt out of the three starts (this postseason) so far."

The Cardinals snapped the Giants' five-game road winning streak in the postseason, three of them this year. Bochy waited until after Game 3 to announce Tim Lincecum as his starter for Game 4 against Adam Wainwright, and also said Barry Zito would start ahead of struggling lefty Madison Bumgarner in Game 5 against Lance Lynn.

Lincecum has allowed one run in eight innings in three relief appearances in the postseason.

"I think we feel that it's time to give Madison a little break," Bochy said of Bumgarner, who allowed six runs in 3 2-3 innings in Game 1.

Beltran is batting .400 in the postseason with three homers and six RBIs, but Carpenter had big numbers against Cain. Before the homer in the third that put St. Louis up 2-1, he was 4 for 4 for his career against Cain, all four singles.

This one was a much bigger deal, a drive that soared over the Cardinals' bullpen in right field and was estimated at 421 feet.

"All those at-bats were a grind facing him, and it was the same way tonight," Carpenter said. "You don't expect things like that to happen."

The 26-year-old Carpenter entered the game 1 for 5 in the postseason, all five pinch-hit appearances, with an RBI single in the wild-card playoff against Atlanta. He got 14 of his 46 RBIs in April as the primary sub at first base for injured Lance Berkman.

On Tuesday, Carpenter was among a group of seldom-used hitters trying to stay sharp by facing Jake Westbrook in a simulated game. The rest of the team had the day off.

Umpires called for the tarpaulin right after the Cardinals made it 3-1 on a run-scoring single by Shane Robinson and Cain was lifted.

It was the third game delayed by rain this postseason and a fourth, Game 4 of the Yankees-Tigers ALCS, was postponed later Wednesday night. Two games between the Yankees and Orioles in Baltimore began late because of inclement weather.

The rain intensified less than 10 minutes after the field was covered, chasing most fans who had remained in their seats to that point. Spotters for the National Weather Service reported 60 mph winds in nearby St. Charles County.

A highlight of the delay was a Pac-Man style chase. Ushers pursued and finally apprehended a fan who jumped out of the stands to get a baseball near the warning track in left field, and then jutted in and out of aisles to elude several ushers who had been closing in.

The storm had been widely anticipated. Some forecasts called for a 70 percent chance of rain. Both managers fielded questions Tuesday and Wednesday about whether the probability of precipitation would affect their selection of the starting pitcher.

The Giants entered 70-22 when scoring first, including the postseason, and took the lead in the third on Pablo Sandoval's run-scoring groundout after leadoff hits by Angel Pagan and Scutaro, whose legs looked just fine on an opposite-field double flared just over first baseman Allen Craig's glove.

Beltran leads all players with eight extra-base hits in the 2012 playoffs and is a career .375 hitter in the postseason, highest ever among players with a minimum of 100 at-bats.

NOTES: Danny Cox, who pitched for Cardinals World Series teams in 1985 and 1987, threw a perfect strike on the first pitch. ... Jay, who was hit by a pitch to start the game, was plunked 15 times in the regular season. ... Matheny had 122 lineups during the regular season but has stuck with the same eight throughout the postseason. ... The Cardinals' win ended a streak of scoring at least six runs in the last eight postseason victories dating to Game 3 of the World Series last year, the longest streak of its kind in postseason history. St. Louis entered averaging 7.6 runs in 16 wins the last two postseasons and just 2.3 runs in the 10 losses. ... Carpenter and Pete Kozma are the first Cardinals rookie teammates to homer in the same postseason history. ... David Freese had two hits for St. Louis and is a career .455 hitter (15 for 33) in the NLCS with four homers, four doubles and 11 RBIs.

Cold viruses point the way to new cancer therapies

Wednesday, October 17, 2012

Cold viruses generally get a bad rap----which they've certainly earned----but new findings by a team of scientists at the Salk Institute for Biological Studies suggest that these viruses might also be a valuable ally in the fight against cancer.

Adenovirus, a type of cold virus, has developed molecular tools----proteins----that allow it to hijack a cell's molecular machinery, including large cellular machines involved in growth, replication and cancer suppression. The Salk scientists identified the construction of these molecular weapons and found that they bind together into long chains (polymers) to form a three-dimensional web inside cells that traps and overpowers cellular sentries involved in growth and cancer suppression. The findings, published October 11 in Cell, suggest a new avenue for developing cancer therapies by mimicking the strategies employed by the viruses.

"Cancer was once a black box," says Clodagh O'Shea, an assistant professor in Salk's Molecular and Cell Biology Laboratory, who led the study. "The key that opened that box was revealing the interactions between small DNA tumor virus proteins and cellular tumor suppressor complexes. But without knowing the structure of the proteins they use to attack cells, we were at a loss for how these tiny weapons win out over much larger tumor suppressors."

O'Shea's team studied E4-ORF3, a cancer-causing protein encoded by adenovirus, which prevents the p53 tumor suppressor protein from binding to its target genes. Known as the "guardian of the genome," p53 normally suppresses tumors by causing cells with DNA damage----a hallmark of cancer----to self-destruct. The p53 tumor suppressor pathway is inactivated in almost every human cancer, allowing cancer cells to escape normal growth controls. Similarly, by inactivating p53, the E4-ORF3 protein enables adenovirus replication in infected human cells to go unchecked.

Two years ago, O'Shea discovered that E4-ORF3 clears the way for adenovirus to proliferate by deactivating genes that help the cell defend itself against the virus. "It literally creates zip files of p53 target genes by compressing them until they can no longer be read," she explains.

E4-ORF3 self-assembles inside cells into a disordered, web-like structure that captures and inactivates different tumor suppressor protein complexes. Horng Ou, a postdoctoral researcher in O'Shea's laboratory, says E4-ORF3 is unusual. "It doesn't resemble any known proteins that assemble polymers or that function in cellular tumor suppressor pathways," he says. "Most cellular polymers and filaments form uniform, rigid chains. But E4-ORF3 is the virus's Swiss army knife----it assembles into something that is highly versatile. It has the ability to build itself into all sorts of different shapes and sizes that can capture and deactivate the many defenses of a host cell."

Dr. O'Shea discusses her research. Credit: Courtesy of the Salk Institute for Biological Studies

In collaboration with scientists from the National Center for Microscopy and Imaging Research at University of California, San Diego, led by Mark Ellisman, the center's director, O'Shea's team used new techniques to reveal the ultrastructure of the remarkable polymer that E4-ORF3 assembles in the nucleus----something that previously had proven difficult since the polymer is effectively invisible using conventional electron microscopy. "What you see is the E4-ORF3 polymer bending and weaving and twisting its way through the nucleus," she says. "It does appear to have a single repeating pattern and creates a matrix that captures several different tumor suppressors and silences p53 target genes."

Initially, E4-ORF3 forms a dimer, made up of only two subunits. In this form, E4-ORF3 largely ignores its cellular targets. The researchers theorized that when E4-ORF3 assembles into a polymer, however, it binds to tumor suppressor targets far more aggressively. To test this theory, they genetically fused E4-ORF3 polymer mutants to lamin, a cellular protein that assembles intermediate filaments that provide stability and strength to cells. They showed that the lamin-E4-ORF3 fusion protein assembled into cylinder-like superstructures in the nucleus that bind and disrupt PML, a protein complex that suppresses tumors.

The Salk findings may help scientists develop small molecules----the basis for the vast majority of current drugs----capable of destroying tumors by binding and disrupting large and complex cellular components that allow cancer cells to grow and spread. Understanding how viruses overcome healthy cells may also help scientists engineer tumor-busting viruses, which offer a new and potentially self-perpetuating cancer therapy. Such modified viruses would destroy only cancer cells, because they could only replicate in cells in which the p53 tumor suppressor has been deactivated. When a cancer cell is destroyed it would release additional copies of the engineered viruses, which would seek out and kill remaining cancer cells that have spread throughout the body.

Engineering these viruses requires disabling the ability of the E4-ORF3 protein to inactivate p53 in healthy cells----otherwise, the virus could destroy healthy cells as well as cancer cells. At the same time, E4-ORF3 has certain important functions in allowing the virus to replicate in the first place, so it can't be completely removed from the virus's arsenal. Thus, the Salk researcher's work on understanding the protein's precise structure, functions and interactions is crucial to engineering viruses in which E4-ORF3's abilities have been precisely modified.

Zenprop gets R151m loan from Nedbank - Commercial, News

Nedbank Corporate Property Finance has approved a loan of R151 million for the expansion of Brandhouse warehouse facilities in Germiston.

In total, the extensions will increase the Brandhouse facilities in Elandsfontein, Germiston by over 24,000 square metres ? the required increase in space resulting from the company?s success and expansion in the South African market.

The loan, which was granted to the Zenprop Group, will fund the development of Brandhouse II, which comprises additional warehouse space for this leading South African premium alcohol beverage company, as well as over 700 square metres of offices and additional hardstand and parking capacity.

In total, the extensions will increase the Brandhouse facilities in Elandsfontein, Germiston by over 24?000 square metres ? the required increase in space resulting from the company?s success and expansion in the South African market.

Brandhouse (Pty) Ltd is one of the leading premium alcohol beverage companies in South Africa offering consumers an unrivalled portfolio of spirits, beer and ready-to-drink (RTD) brands.

The company has been operating in the South African market since July 2004.

In South Africa, Brandhouse operates in four mega-regions, each headed up by a general manager.

These regions are: Western Cape and Port Elizabeth, KwaZulu-Natal and East London, Inland (North) and Inland (South).

Brandhouse currently employs more than 900 permanent employees in South Africa, and is present in all major metropolitan areas as well as many smaller regional centres.

According to Ken Reynolds, regional executive for Nedbank Corporate Property Finance in Gauteng, the decision to grant the loan was not a difficult one to make given the outstanding reputation of the Zenprop Group and the long-standing relationship it has with the bank.

?In addition to the extensive experience of the Zenprop management team, the excellent location of the warehouse development, and a strong, well-established tenant to support the lease,?

Reynolds explains that the group and its various subsidiaries have partnered with Nedbank Corporate Property Finance on numerous successful developments in the past, including similar warehouse facilities and office buildings in Riverhorse Valley and Woodmead Office Park.

Reynolds points out that the latest loan facility reflects Nedbank Corporate Property Finance?s commitment to understanding its clients? business models and industries while partnering with them in the long-term to offer innovative finance options designed to help ensure their sustainable success and growth.

It?s a philosophy that he says lies at the heart of Nedbank Corporate Property Finance?s own success and has played a key role in its achievement of a significant share of the commercial and industrial property finance market across all regions in South Africa.

?The Nedbank Corporate Property Finance strategy has always been absolutely client centric, so we obviously pay due cognisance to the risks and potential of the developments we finance.?

He says a key factor is always the assessment of the people behind the development, which is why they have managed to develop such exceptional partnerships with solid South African property groups like Zenprop, whose values and business practices so closely mirror the bank.

Reynolds points to these strong client partnerships as key to the sustained growth of the Nedbank Corporate Property Finance business during times of economic difficulty.

?By combining this client commitment with an in-depth understanding of the markets in which we operate, Nedbank has achieved a level of robustness and reliability that large, high profile clients like Zenprop really appreciate.?

He adds that this allows the bank to continue to strengthen its relationships with vital commercial and industrial property market stakeholders, not just for the benefit of their businesses, but for the future of the South African property industry as a whole.
